چکیده

The accumulation of sulfate-(35)S by Porphyridium aerugineum cells and subsequent appearance of solubilized capsular polysaccharide-(35)S in the growth medium were examined The uptake of label by the cells was largely light dependent. Pulse-chase experiments using log phase cells revealed a rapid labeling of solubilized capsular polysaccharide, recovered from the medium as the cetylpyridinium chloride precipitate Polyacrylamide gel electrophoresis of the polysaccharide-(35)S showed the sulfate to be firmly bound to an immobile fraction. Sephadex chromatography revealed the molecular weight of the polysaccharide to be in excess of 2 x 10(5). Acid hydrolysis of the polysaccharide-(35)S released sulfate-(35)S ion as evidenced by radioautography of thin layer chromatographs Preliminary electron microscope evidence suggests that the synthesis, movement, and deposition of the capsular polysaccharide on the cell surface are Golgi complex-mediated processes

Precursor-Product Relationships during Sulfate Incorporation into Porphyridium Capsular Polysaccharide.

This study describes the kinetics of (35)S-incorporation during in vivo sulfate esterification of Porphyridium aerugineum capsular polysaccharide. Techniques were developed to isolate the precursor pool (free sulfate), cell-associated product, and extracellular product. Specific radioactivities of these three fractions were monitored during pulse-chase sequences. Label rapidly appeared in the p...

In vivo molybdate inhibition of sulfate transfer to porphyridium capsular polysaccharide.

Active transport of exogenous sulfate into log phase cells of Porphyridium aerueineum followed Michaelis-Menten kinetics, and the apparent Km for sulfate transport is approximately 2.5 x 10(-6)m. Molybdate, also a group VI anion, is a competitive inhibitor of sulfate transport, and the inhibition is freely reversible. Once in the cell, molybdate depresses the rate of sulfate pool utilization by...

The covalent linkage of protein to carbohydrate in the extracellular protein-polysaccharide from the red alga Porphyridium cruentum.

The extracellular anionic polysaccharide isolated from cultures of a unicellular red alga, Porphyridium cruentum, contains a small amount of protein after extensive purification. The polysaccharide and protein are recovered in the same fraction after isopycnic CsCl-density-gradient centrifugation in 4M-guanidinium chloride, under conditions designed to separate proteins from polysaccharide. The...

Relationship between the Unicellular Red Alga Porphyridium sp. and Its Predator, the Dinoflagellate Gymnodinium sp.

Contamination of algae cultivated outdoors by various microorganisms, such as bacteria, fungi, algae, and protozoa, can affect growth and product quality, sometimes causing fast collapse of the cultures. The main contaminant of Porphyridium cultures grown outdoors in Israel is a Gymnodinium sp., a dinoflagellate that feeds on the alga. Comparison of the effects of various environmental conditio...

Expression of E.coli capsular polysaccharide requires the KfiB protein:A Structural based analysis

Abstract Background and objectives: important virulence factor for many invasive bacterial pathogens of humans. Escherichia coli offer a model system to study the mechanisms by which capsular polysaccharides are synthesized and exported onto the cell surface of bacteria. Biosynthesis of the E consists of the repeat structure -4) GlcA- (1, 4)-GlcNAc- (1-, requires the KfiA,...
